THE SAFETY AND EFFICACY OF GH rDNA, LONG TERM TREATMENT IN DISABLED CHILDREN WITH GHD.
THE TREATMENT WITH GH rDNA IN CHILDREN, CHILD AND YOUTH, WITH DELAY OF GROWTH BY GHD HAS BEEN DESIGNED TO DEMONSTRATE THE SAFETY AND EFFICACY OF THE LONG TERM WITH GH rDNA: DR. JOÃO SANTOS CAIO JR. ET DRA. HENRIQUETA VERLANGIERI CAIO.
The treatment with GH rDNA is safe, effective and well tolerated during long term treatment of children with GHD. This was the conclusion reached that a study conducted in Spain: safety and efficacy of GH rDNA in long term treatment of children disabled, child and adolescent with GHD: results of a phase III study. Spanish individuals treatment naïves, prepubertal, child, juvenile and adolescent (n=70) with isolated GHD was treated. Positive change was observed in the height, height standard deviation score (HSDS), the height velocity (HV) HV standard deviation score pattern (HVSDS), serum factor insulin-like growth factor 1 (IGF-1) and insulin-like growth factor deviation score binding protein (IGFBP) shows an increase that treatment gave a good response to growth after 4 years, as demonstrated by a significant increase in mean body height, longitudinal or linear growth (31.1 cm were recorded [95% CI: 29.6 to 32.6]), HSDS (Tunner) (1.42 [1.13 to 1.70]), HV (values 2.4 cm [1.7 to 3.1]), and HVSDS (3.5 [2.7-4.3]). The mean levels of IGF-1 and IGFBP-3 in the serum also increased significantly. Arriving at a correct assessment that correct doses of GH by rDNA is safe and very effective in treating GHD and well tolerated during long term treatment of children, infant and youth with GHD.
The cascade of growth hormone-GH and the remarkable advances over the past decades in our knowledge of its components hormone are considerable. The advent of GH rDNA and their availability for the treatment of a large number of idiopathic GH deficient children, juvenile and adolescent (the minimum rate that prevalence in the U.S. and UK is of 1 in 3.400 and 4.000) dramatically changed image from 1985, the statistics in Brazil prospectively follows approximately the same as U.S.. In the treatment of patients with less than 4 years old, the final height may exceed the height of the target. It is now known that the replacement dose of GH rDNA must be individualized. The earlier made the correction, the better the final result of the expected growth.
